DNA and free radicals looks at the tremendous advances achieved in recent years towards understanding the role of reactive oxygen species in producing DNA damage. The book explores the molecular consequences of severe oxidative stress and its links with DNA damage in cancer, the ageing process, and the pathology of human disease. Discussing the evidence for free radical damage to DNA in cells subjected to oxidative stress, the authors show how such damage can occur and set out the methodology for its measurement. They review the reactions of DNA with various oxygen-derived species, presenting evidence that oxidative damage to DNA in vivo can involve OH- radicals. There is also a review of mechanisms of oxidative stress involving rises in intracellular 'free' Ca2+.The third section of the book is devoted to consequences of oxidative DNA damage —including its relation to cell proliferation and its roles in ornithine decarboxylase regulation and tumour promotion. Enzyme systems that repair damaged DNA are discussed in detail. The final section reviews some of the mechanisms by which oxidative stress occurs, including redox-cycling drugs, the role of mineral fibers and cigarette smoke and the pro-oxidant effects of plant compounds, antioxidant food additives and nutrients.
